安肠汤对腹泻型肠易激大鼠干细胞因子/酪氨酸激酶受体通路的影响
Effect of Anchang Decoction on the SCF/C-kit Pathway in IBS-D Rats

Abstract
Objective:To observe the effect of Anchang Decoction on the stem cell factor/tyrosine kinase receptor(SCF/C-kit)pathway in diarrhea-predominant irritable bowel syndrome(IBS-D)model rats.Methods:Forty male SPF SD rats were randomly and equally divided into normal group,model group,Anchang Decoction group,and Western medicine group.Except for the normal group,IBS-D models were established by capsaicin gavage combined with restraint and tail-clamp stress.After successful modeling,the Western medicine group received pinaverium bromide tablets via gavage,and the Anchang Decoction group received Anchang Decoction via gavage.Hematoxylin-eosin(HE)staining was used to observe pathological changes.Western blot was used to detect protein expression levels of SCF,C-kit,and tumor necrosis factor-α(TNF-α)in colon tissues.RT-PCR was used to detect mRNA expression levels of SCF,C-kit,and TNF-α in colon tissues,and ELISA was used to measure serum TNF-α levels.Results:HE stai-ning of colon tissues showed no abnormal pathological changes in the normal group.In the model group,a small amount of inflam-matory cell infiltration was observed in the colonic mucosa.In the Western medicine and Anchang Decoction groups,occasional in-flammatory infiltration was observed.Compared with the normal group,SCF and C-kit protein and mRNA expression levels in colon tissues of the model group were significantly downregulated(P<0.01),while TNF-α protein and mRNA in colon tissues and serum TNF-α levels were significantly upregulated(P<0.01).In the Western medicine and Anchang Decoction groups,SCF and C-kit protein and mRNA expression levels in colon tissues were upregulated(P<0.01 or P<0.05),while TNF-α protein and mRNA in colon tissues,and serum TNF-α levels were significantly downregulated(P<0.01).Conclusion:Anchang Decoction can improve visceral hypersensitivity in IBS-D model rats,and its mechanism may be related to inhibition of TNF-α expression through regula-tion of the SCF/C-kit signaling pathway.关键词
