身痛逐瘀汤联合骨髓间充质干细胞干预膝骨关节炎大鼠机制研究
Mechanism of Shentong Zhuyu Decoction Combined with Bone Marrow Mesenchymal Stem Cells in Intervention of Knee Osteoarthritis in Rats

Abstract
Objective:To investigate the mechanism by which Shentong Zhuyu Decoction regulates the phosphatidylinositol 3-ki-nase(PI3 K)/protein kinase B(AKT)/nuclear factor κB(NF-κB)signaling pathway in rats with knee osteoarthritis(KOA).Meth-ods:The KOA model was established by collagenase injection and rats were randomly divided into four groups:blank group,control group,Shentong Zhuyu Decoction group,and Shentong Zhuyu Decoction+bone marrow mesenchymal stem cells(BMSC)group.The Shentong Zhuyu Decoction group received Shentong Zhuyu Decoction,the Shentong Zhuyu Decoction+BMSC group received Shent-ong Zhuyu Decoction combined with BMSC,and the control group received saline,all continuously administered for 14 days.At the end of treatment,BMSC proliferation rate and apoptosis index were measured,and hematoxylin-eosin staining was used to observe synovial morphology.Western blot(WB),enzyme-linked immunosorbent assay(ELISA),and real-time quantitative polymerase chain reaction(RT-PCR)were used to detect the expression of Toll-like receptor 2(TLR2),Toll-like receptor 4(TLR4),glyceral-dehyde-3-phosphate dehydrogenase 1(GAPDH-1),glyceraldehyde-3-phosphate dehydrogenase 2(GAPDH-2),alkaline phosphatase 3(AKP3),NF-κB,interleukin-6(IL-6),and interleukin-10(IL-10).Results:Compared with the control group,the Shentong Zhuyu Decoction and Shentong Zhuyu Decoction+BMSC groups showed increased BMSC proliferation rates and reduced apoptosis indices(P＜0.01),with higher proliferation and lower apoptosis in the Shentong Zhuyu Decoction+BMSC group compared with the Shen-tong Zhuyu Decoction group(P＜0.05).Protein expression levels of TLR2 and TLR4 in both treatment groups were significantly lower than those in the control group(P＜0.01),and were lower in the Shentong Zhuyu Decoction+BMSC group than in the Shen-tong Zhuyu Decoction group,though the difference was not statistically significant(P＞0.05).GAPDH-1 and GAPDH-2 protein levels were higher in both treatment groups than in the control group(P＜0.05),with no significant difference between the two groups(P＞0.05).Compared with the control group,AKP3 content in both treatment groups was decreased(P＜0.05,P＜0.01),PI3K and AKT expression was reduced,though differences were not statistically significant(P＞0.05),and NF-κB content in both treatment groups was significantly lower(P＜0.01).Conclusion:Shentong Zhuyu Decoction combined with BMSC may exert thera-peutic effects on KOA model rats by inhibiting the PI3K/AKT/NF-κB signaling pathway,down-regulating the expression of TLR2,TLR4,AKP3,NF-κB,IL-6,and IL-10,upregulating GAPDH-1 and GAPDH-2 protein levels,and enhancing BMSC proliferative ac-tivity.关键词
