续断种子方调控PI3K-Akt-mTOR通路影响少弱精子症模型大鼠精子发生的实验研究
Experimental study on the regulation of PI3K-Akt-mTOR pathway by Xuduan Zhongzi For-mula(续断种子方)and its effect on spermatogenesis in oligoasthenozoospermia model rats

Abstract
Objective Exploring the effect of Xuduan zhongzi Formula(续断种子方,XDZZF)mediated PI3K-Akt-mTOR signaling pathway on spermatogenesis and development in oligoasthenozoospermia model rats.Methods Ten male SD rats were randomly selected as the normal control group.Thirty oligoasthenospermia model rats were established using cyclophosphamide and subsequently divided into the model group,levocarnitine group,and Xuduan Seed Formula group.The Xuduan Seed Formula group received traditional Chi-nese medicine granules via gavage,the western medicine group was administered levocarnitine oral solution,while the normal and model groups received saline.After six weeks,samples were collected.Body weight,testicular index,and pathological scores were evaluated.Sperm concentration and motility were measured,and the expression levels of PI3K-Akt-mTOR pathway-related proteins were detected.Additionally,the micro-and ultrastructural morphology of testicular tissues was analyzed.Results Compared with the model group,the body weight,testicular weight,and testicular index of rats in the Xuduan Seed Formula group were significantly increased(P<0.05).Compared with the normal group,the sperm concentration and motility in the model group were decreased(P<0.01),with extensive apoptosis of spermatogenic cells and disrupted micro-ultrastructural morphology.In contrast,the Xuduan Seed Formula group exhibited significantly higher sperm concentration and motility compared with the model group(P<0.05),along with more regular spermatogenic cell morphology and largely intact micro-ultrastructure.The pathological testicular scores of the Xuduan Seed Formula group were superior to those of both the model group and the L-carnitine group(P<0.05).Furthermore,compared with the model group,the protein expression levels of PI3K,Akt,mTOR,Bax,and Caspase-3 in testicular tissues were significantly reduced in the Xuduan Seed Formula group(P<0.05),while the Bcl-2 protein level was markedly elevated(P<0.05).Conclusion The Xuduan Seed Formula improves sperm quality in oligoasthenospermia rats,potentially by regulating the PI3K-Akt-mTOR pathway,modulating spermatogenic cell apoptosis,and maintaining testicular spermatogenic homeostasis.关键词
