加味升降散调节mTOR/p70S6k通路减轻糖尿病肾脏病肾损伤的机制研究
Mechanism Study of Jiawei Shengjiang San(加味升降散)in alleviating renal injury in diabetic kidney disease by regulating the mTOR/p70S6k pathway

Abstract
Objective To investigate the mechanism of Jiawei Shengjiang San(加味升降散,JWSJS),a modified powdered medicine,in alleviating renal injury in db/db mice with diabetic kidney disease(DKD).Methods 40 db/db mice were randomly divided into a model group,an irbesartan group(0.0195g·kg-1),and the low/medium/high-dose JWSJS groups(1.84,3.68,7.36g·kg-1).The db/m group was the normal control group,with 8 mice in each group,and was intervened continuously for 8 weeks.Fasting blood glucose(FBG),24-hour urinary protein(UTP),and serum BUN,CREA,TG,and CHO levels were measured.Renal pathological changes were evaluated by HE staining,PAS staining,and electron microscopy.Immunohistochemistry(IHC)was used to detect the expression levels of renal podocyte marker proteins,such as Nephrin and Podocin,as well as Beclin1 and LC3.The protein expression levels of LC3,Beclin1,mTOR,p-mTOR,p70S6k,and p-p70S6k in the mice kidneys were determined by Western blot(WB).Results Com-pared with the model group,the FBG,UTP,BUN,CREA,TG,and CHO of mice in the irbesartan group and JWSJS group were signifi-cantly reduced(P<0.05).Renal Pathological damage was alleviated to varying degrees.The expression levels of podocyte marker pro-teins Nephrin and Podocin,and autophagic proteins LC3 and Beclin1,were significantly upregulated(P<0.05).Additionally,the LC3-II/LC3-I ratio and Beclin1 protein levels were significantly increased,whereas the protein ratios of p-mTOR/mTOR and p-p70S6K/p70S6K were significantly decreased(P<0.05).Conclusion JWSJS can alleviate the kidney tissue damage,reduce proteinuria,and ameliorate the kidney function in db/db mice.The mechanism may involve suppression of the mTOR/p70S6k signaling pathway activa-tion and modulation of podocyte autophagy.关键词
