柴胡桂枝汤改善心肌梗死后抑郁大鼠抑郁行为的机制研究

Abstract
Objective To investigate the effects and potential mechanisms of Chaihu Guizhi Decoction(柴胡桂枝汤,CHGZD)on depressive-like behaviors in the model rats of post-myocardial infarction(MI)depression.Methods A rat model of post-MI depression was established using coronary artery ligation combined with behavioral assessments.Model rats were divided into the model group,CHGZD group(7.74 g/kg),and positive control group(fluoxetine 2 mg/kg),with healthy rats as the control group.All groups received daily intragastric administration for 28 consecutive days.Results Compared with the model group,rats in the CHGZD group exhibited:increased sucrose preference;enhanced locomotor activity in the open field test(longer distance traveled,faster speed,increased rear-ing frequency);prolonged immobility latency in the forced swim test(P<0.05,P<0.01);elevated left ventricular ejection fraction(LVEF)and left ventricular fractional shortening(LVFS)(P<0.05);reduced serum levels of CK-MB,BNP,TNF-α,IL-1β,and IL-6(P<0.05);increased levels of serotonin(5-HT),norepinephrine(NE),and dopamine(DA)in the hippocampus(P<0.01);alle-viated myocardial histopathological damage and fibrosis;enhanced mRNA expression of protein kinase B(AKT)and brain-derived neu-rotrophic factor(BDNF)(P<0.05,P<0.01)with decreased mRNA expression of GSK-3β(P<0.05)in the hippocampus;increased protein expression of AKT and BDNF,and decreased protein expression of GSK-3β in the hippocampus.Conclusion CHGZD alleviated depressive-like behaviors in rats with post-MI depression.The underlying mechanism may be associated with activation of the AKT/GSK-3β signaling pathway and upregulation of BDNF protein expression.关键词
