肾脑复元汤通过Mir-210/BCL-2/Beclin-1轴调控缺血性脑卒中细胞自噬的作用机制
Shennao Fuyuan Decoction(肾脑复元汤)regulates autophagy in ischemic stroke via the Mir-210/BCl-2/Beclin-1 axis

Abstract
Objective To investigate the effect of Shennao Fuyuan Decoction(肾脑复元汤,SNFYD)on cellular autophagy after isch-emic injury in focal cerebral ischemia rats and its regulatory role in cerebral ischemia.Methods A rat model of focal cerebral ischemia(MCAO)was established.Thirty-six male SD rats were randomly divided into three groups:the blank control group,model group,and SNFYD group.Tissues were harvested after 3 days of administration.Brain tissue structure was observed by HE staining.The expres-sions of microtubule-associated protein 1 light chain 3(LC3),beclin-1,and B-cell lymphoma-2(BCL-2)were detected by Western blot(WB).LC3 expression was detected by immunofluorescence.The mRNA expressions of LC3,Beclin-1,BCL-2,and microRNA-210(Mir-210)were measured by RT-qPCR.Results Compared with the blank control group,the model group showed increased patho-logical damage,decreased BCL-2 expression(P<0.01),upregulated LC3 and Beclin-1 protein and mRNA levels,and elevated Mir-210 mRNA expression(P<0.01).In contrast,when compared with the model group,the SNFYD group exhibited alleviated tissue damage,downregulated LC3,Beclin-1,and Mir-210 expressions,and upregulated BCl-2 expression(P<0.01).Conclusion SNFYD may exert a protective effect on brain tissue by inhibiting Mir-210 levels,downregulating Beclin-1,upregulating BCL-2,and suppress-ing cellular autophagy.关键词
