阿拉尔垦区二斑叶螨抗药性监测与机制分析
Monitoring and mechanism analysis of resistance in Tetranychus urticae field population from Alar

Abstract
As a worldwide agricultural mite,Tetranychus urticae has developed resistance to a wide range of acaricides.In this study,the resistance level of the Alar strain(Ale)was monitored and the mechanism of drug resistance was analysed,in order to provide a basis for manage it.Toxicological assays showed that compared with Laboratory susceptible strain of T.urticae(Lab-S),the resistance ratio of Ale to cyflumetofen,pyridaben,abamectin and etoxazole were 48.31,94.28,572.10 and over 27 000 times,respectively;the resistance ratios of Ale to amitraz,propargite,chlorfenapyr,λ-yhalothrin,azocyclotin,diafenthiuron,bifenazate,bromopropylate and spirodiclofen ranged from 1.77-13.83;the LC50 of phoxiphos for both populations were greater than 5 000 mg·L-1.Mutations frequencies of Ale's PSST homologue of NADH dehydrogenase(PSST)H92R,chitin synthase 1(CHS1)I1017F,glutamate-gated chloride channel 1(GluCl1)G314D and glutamate-gated chloride channel 3(GluCl3)G326E were 94.07%,100%,66.48%and 86.53%,respectively,while,these mutations weren't detected in Lab-S;mutation frequencies of acetylcholinesterase(AChE)F439W in Lab-S and Ale were 89.19%and 80.98%,respectively,and those of AChEG228S were 84.24%and 61.67%,respectively.There was no significant difference in AChE mutation frequencies between the two populations.In addition,Transcriptome analysis showed significant over-expression of several genes encoding detoxification enzymes in Ale,including 16 cytochrome P450,10 esterase,17 glutathione S-transferase,22 uridine diphosphate glucuronosyltransferase and 8 ABC transporter,which was an important reason for Ale to develop multi-resistance.Therefore,these results indicated that Ale had multiple target genes mutations and up-regulated expression of detoxification enzyme genes,resulting in medium or above resistance to a variety of acaricides.it was recommended to use acaricides with high toxicity and low resistance level alternately or in combination to control T.urticae in Alar,such as amitraz,propargite,azacyclotin,bifenazate and spirodiclofen.关键词
