Study on the effect of NtWo gene on epidermal hair development in tobacco

Abstract
[Objective]This study aims to explore the effect of the NtWo gene on epidermal hair development in tobacco.[Methods]Based on homologous cloning,sequence characteristics,and tissue specific expression pattern analysis of NtWo gene,knockout and overexpression materials of the gene were generated.Glandular hair phenotypes were observed,and changes in the expression levels of genes related to glandular hair formation in the knockout and overexpression materials were analyzed.[Results](1)Two homologous NtWo genes,NtWo-1 and NtWo-2,were identified in tobacco,with coding sequences of 2052 bp and 2121 bp,respectively.The encoded proteins contained HD-LZ,START,and SAD domains,belonging to the HD-ZIP IV subfamily.(2)Subcellular localization results showed that NtWo protein was localized in the nucleus.(3)qRT-PCR analysis showed that both NtWo-1 and NtWo-2 were most highly expressed in epidermal hairs,and NtWo-1 expression was higher than NtWo-2 in all tissues except roots.(4)Overexpression of NtWo-1 significantly increased the density of long-stalked glandular hairs on tobacco leaves,while knockout of the NtWo gene significantly reduced the density of long-stalked glandular hairs.The densities of short-stalked glandular hairs and protective hairs did not change significantly in either overexpression or knockout plants.(5)qRT-PCR results showed that NtWo significantly affected the expression of glandular hair formation-related genes,including NtCycB2,NtHD8,NtHD9,NtHD12,NtMIXTA,NtWox3b,NtLFS,and NtBRC2a.[Conclusion]The NtWo gene positively regulates the formation of long-stalked glandular hairs in tobacco.关键词
