Abstract

Carbonic anhydrase is an enzyme that efficiently catalyzes the hydration of CO2.To overcome its drawbacks of easy deactivation and poor recyclability,carbonic anhydrase-producing bacteria were immobilized and studied for enzyme activity properties.Bacteria capable of producing carbonic anhydrase were isolated from Karst soil and identified through 16S rDNA sequencing and phylogenetic analysis as belonging to the genus Stenotrophomonas sp.Using sodium alginate-chitosan composite materials as carriers,the enzyme activity recovery ratio before and after immobilization was used as an indicator of immobilization effectiveness.Experimental results showed an enzyme activity recovery rate of up to 55%.Response surface methodology was applied to explore the effects of immobilization time,sodium alginate concentration,chitosan concentration,and calcium chloride concentration on enzyme activity recovery rate,revealing the optimal immobilization conditions:2 hours of immobilization time,2%sodium alginate concentration,1%chitosan concentration,and 1 mol·L-1 calcium chloride concentration.It is shown that under these optimal conditions,the stability of the immobilized strain was significantly improved,and reusability was optimized.
