利用单细胞转录组揭示牛窦卵泡发育微环境异质性及动态调控网络
Single-cell transcriptomic profiling reveals heterogeneity and dynamic regulatory networks of the microenvironment during antral follicular development in bovine

Abstract
The development of antral follicles involves multilayered mechanisms,and the research in this field is constrained by the inefficient capture of the physiological follicular microenvironment and key molecular events.This study employed single-cell RNA sequencing(scRNA-seq)to construct a single-cell transcriptional atlas of 3-8 mm follicles,which are commonly used for in vitro embryo production in bovine.Molecular features,including cellular heterogeneity,differentially expressed genes,developmental trajectories,and cell communication,were further investigated.The results revealed that six distinct cell types—granulosa cells,stromal cells,theca cells,smooth muscle cells,endothelial cells,and immune cells[the cell markers are anti-Müllerian hormone(AMH),decorin(DCN),steroidogenic acute regulatory protein(STAR),actin alpha 2,smooth muscle(ACTA2),von Willebrand factor(VWF),CD14]comprise the microenvironment of the antral follicle in bovine,with granulosa cells emerging as the dominant population driving folliculogenesis.The four subclusters of granulosa cells exhibited high heterogeneity[the cell markers are cytochrome P450 family 19 subfamily A member 1(CYP19A1),POU class 5 homeobox 1(POU5F1),centromere protein F(CENPF),and fibroblast growth factor 10(FGF10)]demonstrated a differentiation trajectory progressing through"signal transduction→transcriptional regulation→proliferative division→functional maturation".This trajectory,coordinated with activation of inhibin/inhibin beta A(INH/INHBA)and other immunoregulatory pathways,remodels the follicular microenvironment and initiates the follicle selection program.Furthermore,this study provides molecular evidence at the single-cell resolution for the differentiation pathways between the inner and outer layers of theca cells and identifies the presence of lymphatic endothelial cells.Our results provide novel insights into regulatory mechanism of bovine antral follicular development and offer new strategies for optimizing in vivo and in vitro embryo production systems in bovine.关键词
