产高活性壳聚糖酶菌株CJB-H6的筛选、鉴定及其酶学性质
Screening and Identification of Strain CJB-H6 Producing High-Activity Chitosanase and Enzymatic Properties of Its Chitosanase

Abstract
[Objective]This study aims to screen a high-yield chitosanase-producing strain and investigate the enzymatic properties of the chitosanase produced by this strain,as well as optimize the process conditions for the efficient preparation of chitosan oligosaccharides.[Methods]High-yield chitosanase strains from the sediments in the Yanwei Port area of Lianyungang were screened using the transparent zone method and enzyme activity assay;and chitosanase was purified by fractional precipitation and chromatography,and its enzymatic properties were characterized;single-factor experiments were conducted to optimize the conditions for the efficient production of chitosan oligosaccharides by chitosanase,using hydrolysis rate and oligosaccharide yield as indicators.[Results and Conclusion]Bacillus sp.CJB-H6 with chitosanase activity of(245.22±3.18)U/mL was obtained.The purified chitosanase from Bacillus sp.CJB-H6(Bpt CSN)had a molecular mass of approximately 34 kDa and a specific activity of(9 889.33±47.21)U/mg.Its optimal catalytic temperature and pH were 55℃and 6.0,respectively.Metal ions Ba2+and Mg2+significantly enhanced the activity of Bpt CSN(P<0.05).Under the conditions of 55℃,the optimal process conditions for Bpt CSN catalyzing 60 g/L colloidal chitosan are as follows:enzyme amount of 150 U/mL,acetic acid volume fraction of 2.0%,stirring speed of 100 r/min,and hydrolysis time of 2.5 hours,thereafter,the hydrolysis rate and oligosaccharide yield reached 94.69%±5.38%and 91.06%±4.91%,respectively.The hydrolysis products are mainly composed of chitobioses,chitotrioses,and chitotetraoses.关键词
