基于环境DNA的北运河流域河川沙塘鳢靶向监测
Targeted monitoring of Odontobutis potamophila in Beiyun River Basin using environmental DNA technology

Abstract
Odontobutis potamophila,as an alien species,is widely distributed in the Beiyun River Basin.Its targeted monitoring plays a crucial role in clarifying the distribution of Odontobutis potamophila and its impact on the aquatic ecosystem of the Beiyun River Basin.Based on environmental DNA(eDNA)quantitative technology,a targeted monitoring method for specific species was established through real-time quantitative polymerase chain reaction(qPCR)detection.Specific primers and probes were designed and screened,and their PCR amplification primer information,reaction conditions,limit of detection,stability,and accuracy were analyzed.The results showed that:the upstream primer was"TAACCATATATACCAGGATTCAACACAC",the downstream primer was"GGAATAATATCAATCAATGGCCCTGAAA",and the probe was"GAGTCAAGCGTTGGAACTA TGTTTCTTGAAGG".The limit of detection was 1 × 101 Copies/μL,and the cycle threshold(CT)value forjudging positive sample detection was 32.1.The method had high stability,and the plasmid at the lowest concentration could be stored at-20 ℃ for at least 7 days.The positive accuracy proportion was about 75%,and the negative accuracy ratio was 100%.Odontobutis potamophila in the Beiyun River Basin had become one of the main fish species in the Beiyun River Basin.These research findings provided support for the conservation and restoration of aquatic ecology in water areas..关键词
