Abstract
OBJECTIVE 1.Compare the differences in enterotoxin virulence genes carried by Staphylococcus aureus in the nasal cavity of patients with chronic rhinosinusitis with nasal polyps(CRSwNP)and chronic rhinosinusitis without nasal polyps(CRSsNP),and explore the relationship between infection with strains carrying different enterotoxin virulence genotypes and the incidence of sinusitis and nasal polyps.2.Clarify the dominant bacterial types of Staphylococcus aureus in the nasal cavity of CRSwNP/CRSNP patients,and provide a basis for prevention and treatment in this region.METHODS 1.Isolation and identification of Staphylococcus aureus:The nasal mucosal surface secretions obtained from the control group and experimental group(including CRSwNP group and CRSsNP group)patients under nasal endoscopy will be cultured,and the positive cultured strains will be identified in the VITEK 2 fully automated microbial identification system.2.Detection of virulence factors:PCR method was used to detect 6 enterotoxin virulence genes carried by 41 isolated and identified strains of Staphylococcus aureus.3.MLST typing:Seven housekeeping genes of Staphylococcus aureus amplified by PCR method were sequenced,and the sequences of the seven housekeeping genes were concatenated.MEGA6.0 software was used for sequence analysis to construct a genetic evolutionary tree.RESULTS The detection rate of seb in the experimental group was significantly higher than that in the control group,the detection rate of seb in the CRSwNP group was higher than that in the CRSsNP group,and the other 5 kinds of enterotoxin genes were not statistically significant between the experimental group and the control group;Staphylococcus aureus adhesin in the experimental group and the control group were detected,and the detection rate was higher,detection rate were not statistically significant between the experimental group and the control group;The detection rate of HLα and HLβ genes encoding hemolysin was higher in the experimental group and the control group,and there was no significant difference between the 2 groups in the experimental group and the control group.There were 16 different ST types which were found in 41 strains of Staphylococcus aureus,and ST59 was the main sequence type.CONCLUSION There is a correlation between Staphylococcus aureus carrying enterotoxin genes and the onset of CRS,and the more strains that may carry enterotoxins,the greater the relationship between infection and nasal polyps.The dominant strain of Staphylococcus aureus in this area is ST59.关键词
