光照与培养基对新型兰种子非共生萌发与生长的影响
Effects of light condition and culture medium on seed non-symbiotic germination and growth of Neogyna gardneriana

Abstract
[Objective]Investigating the effects of light conditions and culture medium types on the asymbiotic germination and seedling growth of Neogyma gardneriana seeds,to establish a foundation for the resource conservation,artificial propagation,and utilization of Neogyna gardneriana.[Method]Using seeds of Neogyna gardneriana as experimental material,they were inoculated into three different culture medium and cultivated for 300 days under either 24-hour light or 24-hour dark conditions.Germination,differentiation,and seedling growth were observed and recorded.[Result]Under all culture conditions,seeds of Neogyna gardneriana began to germinate within 20-36 days,with germination generally occurring earlier under light culture than under dark culture.With the exception of the MS medium under 24-hour dark culture,all other treatments completed the developmental stages from embryo expansion and protocorm formation to differentiation into leaves,roots and pseudobulbs,ultimately establishing seedlings.The highest total germination rate(81.85%)and differentiation rate(69.79%)were observed in the 1/2 MS medium under 24-hour light culture.Seedlings cultured in Hyponex No.1 medium,whether under light or dark conditions,showed better growth than those in other treatments,with the most pronounced improvement in root growth.Two-way ANOVA results indicated that light significantly promoted seed germination,while the culture medium played a major role in differentiation,seedling establishment,and subsequent growth.[Conclusion]The optimal method for promoting seed germination and growth of Neogyna gardneriana involves initial cultivation on 1/2 MS medium under 24-hour light conditions to complete seed germination,differentiation,and seedling establishment,followed by transfer to Hyponex No.1 medium for root development and seedling strengthening.关键词
