Effects of sweet potato puree additives on the proliferation and growth of Dendrobium protocorms

Abstract
[Objective]Investigated the effects of different types of sweet potato puree additives on the proliferation and growth of Dendrobium nobile and D.heterocarpum protocorms,in order to screen out the suitable culture media for the proliferation and growth of Dendrobium protocorms.[Method]Using D.nobile and D.heterocarpum as materials,their protocorms were cultured in media supplemented with three types of sweet potato puree,they were orange-fleshed sweet potato puree,purple-fleshed sweet potato puree,and high-starch sweet potato puree,respectively.The growth of the protocorms were compared with the blank control group and potato puree group,and the proliferation coefficient and chlorophyll content were statistically analyzed.[Result]When a high proportion(≥20%)of sweet potato puree was added,the growth of Dendrobium protocorms were mostly inhibited.After 25 days of culture,Among all treatments,the proliferation coefficient of D.nobile protocorms reached 9.41 in the addition of 10%starch mashed medium,and the chlorophyll content with 20%purple-fleshed sweet potato yielded the highest chlorophyll content of 0.0920 mg/g.The proliferation coefficient of D.heterocarpum protocorms was 7.20 when adding 10%high-starch sweet potato puree,which was 120.76%higher than the blank control group.[Conclusion]Among the tested additives,10%high-starch sweet potato puree was identified as the most suitable for the proliferation and growth of both Dendrobium protocorm types,demonstrating a significant promoting effect.The optimal culture medium for the proliferation of two Dendrobium protocorm types was 1/2 MS+1.0 mg/L 6-BA+0.1 mg/L NAA+10%high-starch sweet potato puree.关键词
