室内海水青鳉循环水养殖系统中细菌群落结构的高通量测序分析
High-throughput sequencing analysis of bacterial community structure in recirculating aquaculture system for Oryzias melastigma

Abstract
[Objective]This study focused on the structure and function of bacterial communities in the recirculating aquaculture sys-tem for Oryzias melastigma to provide basis for the development of healthy marine aquaculture.[Method]Water samples were col-lected from the breeding tank,outlet and sedimentation tank of recirculating aquaculture system to analyze their bacterial community structure and diversity by 16S rDNA V3-V4 high-throughput sequencing.[Result]Bacterial abundance and diversity were similar between the breeding tank and the outlet,while those in the sedimentation tank were significantly decreased.Proteobacteria,Bacte-roidota,Chloroflexi and Planctomycetota made up the main phyla in the recirculating aquaculture system.Specifically,the relative a-bundance of Proteobacteria was significantly higher in the sedimentation tank than those in the breeding tank and at the outlet,while Bacteroidetes and Chloroflexi exhibited the opposite trend.At the genus level,the dominant species at each sampling point was dif-ferent,with Methylotenera,mostly related to functions of nitrogen fixation and phosphorus removal,being the dominant species in the breeding tank,and some genus of Caulobacteraceae and Ralstonia being the dominance at the outlet and in the sedimentation tank,respectively.PICRUSt function prediction revealed that the tertiary functional layer was mainly composed of ABC2 transporter,iron complex outer membrane receptor protein,flagellin and other molecular functional components,and the differences among sam-pling points were significant.[Conclusion]Bacterial community structure of different functional zones in the recirculating aquaculture system was significantly different,and that of outlet and sedimentation tank were more similar.After passing through the biofilter,the diversity of bacterial community decreased significantly and its function also changed significantly.关键词
