华南沿海3种常见牡蛎的GDF-8基因克隆与表达分析
Molecular cloning and expression analysis of GDF-8 gene of 3 common oyster species in south China coastal areas

Abstract
[Objective]This study investigated the expression patterns and function of the GDF-8 gene in Crassostrea hongkongensis,Crassostrea rivularis,and Crassostrea sikamea,to provide a theoretical foundation for unveiling oyster muscle development and main-tenance.[Method]The core coding sequences(CDS)of GDF-8 gene from 3 kinds of oyster were obtained by polymerase chain reac-tion(PCR),and followed by bioinformatics analysis.The expression patterns of GDF-8 in different tissues were detected by real-time fluorescence quantitative PCR(qRT-PCR).[Result]The CDS of GDF-8 gene in the 3 oyster species were 1 317 bp in length,and encoded 438 amino acids.The encoded GDF-8 protein all included TGF-β superfamily domain,and shared homology of 95.48%-96.83%with the GDF-8 of C.gigas.The expressions of ChGDF-8,CarGDF-8 and CsiGDF-8 were the highest in the labrum,which were consecutively declined in the adductor muscle,digestive gland,mantle and gills of C.hongkongensis.The expression of CarG-DF-8 was higher in the mantle of C.rivularis and lowest in the gill.The CsiGDF-8 was highly expressed in the gill of C.sikamea,and least expressed in the adductor muscle.[Conclusion]GDF-8 was involved in the development and maintenance of muscle in adult oysters.关键词
