摘要

Abstract

The STING pathway is a critical signaling pathway in the innate immune system,triggered by cyclic dinucleotides to induce antimicrobial immune responses.Beyond its role in host defense,STING activation also significantly contributes to the regulation of anti-tumor immune re-sponses.Consequently,STING agonists are extensively utilized in research targeting pathogen infec-tion and tumor immunotherapy.However,natural STING agonists exhibit poor stability and limited cell membrane permeability,underscoring the importance of developing novel agonists and efficient delivery systems.This review summarizes recent advances in the design of STING agonists and their delivery strategies,aiming to provide new perspectives for the clinical application of STING pathway modulation in immunotherapy.
