Isolation and growth-promoting properties of plant growth promoting rhizobacteria from Taxus×media rhizosphere

Abstract

Took the rhizosphere soil of Taxus×media as the research object and isolated and identified plant growth-promoting rhizobacteria(PGPR)that can promote its growth.Strains with growth-promoting effects were screened through growth-promoting experiments such as phosphorus solubilization,potassium solubilization,siderophore production,indole acetic acid(IAA)production,based on the wheat germination experiments and Taxus×media cutting experiments,identified specific PGPR strains with significant growth-enhancing effects on Taxus×media.The results showed that,among the 13 strains with different morphologies screened from the rhizosphere soil of Taxus×media,the WC strain possesses the ability to solubilize potassium;the W1,W7,W8,WA,WB,WC,WD and WE strains could produce siderophores;the W3,W7,W8 and WB strains could produce IAA.The results of the wheat germination experiment showed that the screened growth-promoting bacteria had significant growth-promoting effects on the plant height of wheat.The growth-promoting effects of the W1,W8,WA and WB strains were more obvious,but only the WC strain had a good promoting effect on root length.The results of the Taxus×media cutting experiment showed that the W1,WA,WC,WD strains the mixed bacteria of WB and WC had a significant promoting effect on the growth of new branches,the mixed bacteria of WB and WC had a significant promoting effect on the growth of bud scales.Based on the above experimental results,the 16S rDNA of the W1,W8,WA,WB,WC and WD strains were sequenced,which were Exiguobacterium acetylicum,Bacillus mobilis,Bacillus albus,Lysinibacillus sphaericus,Lysinibacillus fusiformis and Escherichia coli respectively.The study provides reliable microbial resources for the research and development of Taxus×media microbial fertilizers and reference basis for Taxus×media cultivation.
