低氧环境诱导外泌体源性miR-141-3p调控滋养细胞生物学功能的研究

Abstract
Objective:To study that the hypoxic environment induces the release of miR-141-3p from exosomes and regulates the biological function of trophoblast cells,as well as to provide a theoretical basis for further studies on the role of miR-141-3p in the pathogenesis of preeclampsia.Methods:Human chorionic trophoblast cells HTR-8SVneo were cultured in vitro and divided in-to a normoxic group and a hypoxic group.The proliferative activity of the cells in each group was assessed by using the CCK-8 as-say and clone formation assay;the invasive and migratory abilities of the cells in each group were assessed by using the Transwell assay.Western blot was used to detect the presence of exosomes.Transmission electron microscopy was used to visualize the mo-lecular morphology of the isolated exosomes.The particle size and concentration of exosomes were detected by nanoparticle track-ing analyzer.qRT-PCR was used to detect the expression level of miR-141-3p in each group of cells and exosomes.Results:Com-pared to the normoxic group,the miR-141-3p expression in HTR-8/SVneo cells and exosomes was elevated,and the proliferative activity and migration and invasive abilities of the cells in the hypoxic group were decreased,while the miR-141-3p expression was decreased,the proliferative activity was increased,and the number of invasive cells was increased in the normoxic group(P<0.01);Western blot showed a strong positive expression of exosome marker proteins.Conclusion:The expression level of miR-141-3p in exosomes is up-regulated under hypoxic conditions,inhibiting cell proliferation and invasive and migratory abilities.Exosome-derived miR-141-3p can regulate the biological functions of trophoblast cells.关键词
