IGF-1调控PI3K/AKT信号通路改善早产大鼠视网膜病变的作用
IGF-1 regulates PI3K/AKT signaling pathway to ameliorate retinopathy of prematurity in rats

Abstract
Objective:To investigate the effect of IGF-1 on phosphatidylinositol 3-kinase-protein kinase B(PIi3k/AKT)sig-naling pathway in intrauterine growth restriction retinopathy of prematurity rats.Methods:SD pregnant rats were randomly divid-ed into the control group,the model group and the treatment group,the serum factor level was detected by ELISA.The 14-day-old rats were used as the research objects.The young rats with retinopathy were selected by ERG,IGF-1 15 μg/mL 0.1 mL was injected into the retina of the model group.The pathological changes of the retina were observed by H&E staining,and the VEGF protein in retinal tissue was detected by immunohistochemistry.The expressions of IGF-1,IGF-1R and VEGF mRNA were detected by RT-PCR,the expression of IGF-1R,VEGF,p-PI3K,PI3KT,p-AKT and AKT were detected by Western blot.Results:The results showed that compared to the control group,there were significant differences in the number of newborn mice,the rate of stillbirth,the rate of deformity,the amplitude of GH and IGF-1 of wave A and B in the model group,the ar-rangement of cells in the model group were disordered after H&E staining in the IGF-1 treatment group,the number of cells de-creased and the retina became thinner.The expression of VEGF in the retina of IGF-1 treated group was lower than that of the model group.The results of RT-PCR showed that the expression of IGF-1 and IGF-1R mRNA in the model group was lower than the control group,while the expression of VEGF mRNA was higher in the treatment group than that in the model group,the mRNA expression in the treatment group was lower than that in the model group(P<0.05).Compared to the control group,the expression of VEGF,p-PI3K/PI3KT and p-AKT/AKT in the retina of the model group was significantly increased,while the ex-pression of IGF-1R was significantly decreased,the expression of VEGF,p-PI3K/PI3KT and P-AKT/AKT were significantly decreased and the expression of IGF-1R was significantly increased in the treatment group(P<0.05).Conclusions:IGF-1 may ameliorate retinal injury in IUGR rats by inhibiting PI3k/AKT signaling pathway.关键词
