Effect of SAA1 on epithelial-mesenchymal transition and stemness properties of esophageal squamous cell carcinoma cells

Abstract
Objective:To explore the effect of serum amyloid A1(SAA1)on epithelial-mesenchymal transition(EMT)and stemness properties of esophageal squamous cell carcinoma(ESCC)cells.Methods:The expression of SAA1 mRNA and cell stemness in ESCC tissues were analyzed using the NCBI-GEO database,and TCGA database was used for survival analysis,dif-ferentially expressed gene analysis and enrichment analysis.Overexpression or knockdown of SAA1 in ESCC cells was performed using SAA1 overexpression lentivirus or SAA1 small interfering RNA(siRNA).Transcriptome sequencing was used to screen differentially expressed genes.Western blot was used to detect the expression of EMT-related proteins and stemness markers.Tumorsphere-formation assay was used to detect the effect of SAA1 on the sphere-forming ability of ESCC cells.Results:SAA1 mRNA expression in ESCC tissues was significantly higher than that in normal tissues.High SAA1 expression indicated a poor prognosis.EMT-related genes were enriched in both the TCGA dataset and the transcriptome sequencing data of SAA1 overex-pressing ESCC cells.Overexpression of SAA1 upregulated the expression of the EMT-related proteins N-cadherin,Vimentin,Snail and Twist1,as well as the stemness marker CD44,and downregulated the expression of E-cadherin.Knockdown of SAA1 had the opposite effects.Overexpression of SAA1 enhanced the sphere-forming capacity of ESCC cells.Conclusion:SAA1 is sig-nificantly upregulated in ESCC tissues and promotes EMT and stemness properties of ESCC cells.关键词
