Experimental study of Panax notoginseng saponins on improving spermatogenic function in mice with depression-like behavior

Abstract
Objective:To explore the effect of Panax notoginseng saponin(PNS)on spermatogenesis in mice with depression-like behavior and its possible mechanism.Methods:A total of 40 male C57BL/6J mice were randomly divided into the Control group and the model group(Complete Freund's Adjuvant,CFA).The depression-like behavior model was induced by injecting CFA into the sole of the right hind paw of mice in the model group.After the establishment of depression-like behavior model,the mice were divided into the model group(CFA-D group)and the drug intervention group(CFA-D+PNS group).Open field ex-periment was used to detect the activity of mice,sugar spray experiment and forced swimming experiment were used to detect depression-like behavior of mice.Testes and epididymis weight,testes coefficient,epididymis coefficient,and sperm count were detected to check reproductive organs and sperm count of mice.Morphological changes of testicular spermatogenic cells and sperm deformities were observed by H&E staining.The changes of serum testosterone(T),luteinizing hormone(LH),gonadotropin re-leasing hormone(GnRH),and follicle stimulating hormone(FSH)were detected by ELISA assay.Results:Compared to the Control group,the activity time(P<0.001)and activity distance(P<0.001)in the model group were significantly reduced,the back licking time was reduced(P<0.001),and the immobile time of forced swimming was prolonged(P<0.01).Testicular weight(P<0.001),epididymal weight(P<0.01),testicular coefficient(P<0.001),epididymal coefficient(P<0.001)and sperm count(P<0.001)were all decreased,and the malform rates of sperm head(P<0.001),neck(P<0.05)and tail(P<0.001)were significantly increased.The number of spermatogenic epithelial cells decreased and the expression of T(P<0.01),FSH(P<0.001),GnRH(P<0.001),LH(P<0.001)in serum decreased significantly.After PNS intervention,the inner frame activity time(P<0.01)and activity distance(P<0.001)were significantly increased,the back licking time was extended(P<0.001),and the immobile time of forced swimming was shorter(P<0.01).Testicular weight(P<0.001),epididymal weight(P<0.01),testicular coefficient(P<0.001),epididymal coefficient(P<0.05),and sperm count(P<0.01)were signifi-cantly increased,and the malform rates of sperm head(P<0.01),neck(P<0.05),and tail(P<0.01)were significantly de-creased.The spermatogenic epithelial cells were closely arranged and increased in number.Meanwhile,serum T(P<0.05),FSH(P<0.001),GnRH(P<0.01),LH(P<0.001)were increased.Conclusion:PNS can significantly improve reproductive organ atrophy and spermatogenic dysfunction in depression-like mice.At the same time,the concentrations of T,LH,GnRH,and FSH in serum were increased,thus improving spermatogenic function of depression-like behavior mice.关键词
