Effect of electroacupuncture on orexin receptor expression in PVT brain region of chronic sleep deprivation rats

Abstract
Objective:To observe the regulatory effects of electroacupuncture on orexin receptor 1(OX1R)and orexin recep-tor 2(OX2R)in the paraventricular thalamus(PVT)of rats with chronic sleep deprivation(CSD),and to analyze the mechanism by which electroacupuncture improves CSD rats.Methods:A total of 48 male SD rats were randomly divided into four groups us-ing the random number method:the blank group(Control),the model group(CSD),the electroacupuncture group(CSD+EA),and the sham acupuncture group(CSD+SA)with 12 rats in each group.The modified multi-platform method(MMPM)was employed to establish the CSD rat model.In the electroacupuncture group,electroacupuncture interventions were carried out at"Baihui"(GV20),"Shenmen"(HT7),"Neiguan"(PC6),and"Taichong"(LR3)for 14 days.The electroencephalogram(EEG)and electromyogram(EMG)detection techniques were utilized to observe the sleep differences among the rats.Real-time fluorescence quantitative polymerase chain reaction(qPCR)and Western blot assays were used to detect the expression levels of OX1R and OX2R mRNA and proteins in the PVT brain region of the rats.Results:In the CSD model group,the time during the wakefulness period(WAKE period)of the rats was significantly increased,while the cumulative times during the non-rapid eye movement sleep period(NREM period)and the rapid eye movement sleep period(REM period)were decreased(P<0.001,P<0.01).The mRNA transcription level and protein expression level of OX1R in the PVT both exhibited a significant upward trend(P<0.01,P<0.001).After electroacupuncture treatment,the sleep-wakefulness balance of the CSD rats was restored.The cu-mulative time during the WAKE period was significantly reduced,and the NREM period,REM period,and total sleep time were all significantly prolonged(P<0.001,P<0.01).The expression of OX1R at the mRNA and protein levels in the PVT was de-creased(P<0.01,P<0.001).There was no significant difference in the expression of OX1R and OX2R in the PVT between the sham acupuncture group and the model group(P>0.05).Conclusion:Electroacupuncture can effectively regulate the sleep-wake rhythm and anxiety state of CSD rats,and its mechanism may be related to the inhibition of the expression of orexin receptors in the PVT brain region of CSD rats.关键词
