Study on Curcumol Inhibiting Inflammatory Factor Secretion,Proliferation and Apoptosis of Endometrial Stromal Cells by Intervening in JAK2/STAT3 Pathway

Abstract
Objective:To investigate the effect of curcumol on inflammatory factor secretion,proliferation,and apoptosis of endo-metrial stromal cells(ESCs)through regulating the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)pathway.Methods:Six endometrial tissues were collected respectively from patients with normal endometrium and those with endometri-osis,followed by isolation and culture of endometrial stromal cells.The cells were divided into four groups:normal endometrial stromal cells(NESCs)group,ectopic endometrial stromal cells(EESCs)group,AG490 group(EESCs cultured with AG490,a JAK2/STAT3 pathway inhibitor),and curcumol group.Cell Counting Kit-8(CCK-8)was used to determine the proliferative activity of endometrial cells;flow cytometry to analyze cell apoptosis;quantitative real-time polymerase chain reaction(qRT-PCR)to detect the mRNA ex-pressions of cysteinyl aspartate-specific proteinase 3(Caspase 3),B-cell lymphoma 2(Bcl-2),and Bcl-2-associated X protein(Bax,a pro-apoptotic protein of the Bcl-2 family)in cells of each group;enzyme-linked immunosorbent assay(ELISA)to measure the con-tent of inflammatory factors secreted by cells;Western blot to detect the protein phosphorylation levels of Janus kinase 2(JAK2)and signal transducer and activator of transcription 3(STAT3)in cells of each group.Results:Compared with the NESCs group,the EE-SCs group showed a decreased cell apoptosis rate,enhanced proliferative activity,increased expression level of the apoptosis-related gene Bcl-2,decreased expression levels of Bax and Caspase 3,increased secretion of the inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6),upregulated phosphorylation levels of JAK2 and STAT3(p-JAK2/JAK2,p-STAT3/STAT3),and enhanced activity of the JAK2/STAT3 signaling pathway.Compared with the EESCs group,the curcu-mol group exhibited an increased cell apoptosis rate,reduced proliferative activity,decreased expression level of Bcl-2,increased ex-pression levels of Bax and Caspase 3,reduced secretion of TNF-α,IL-1β,and IL-6,downregulated phosphorylation levels of JAK2 and STAT3,and diminished activity of the JAK2/STAT3 signaling pathway.Conclusion:Curcumol can inhibit the JAK2/STAT3 signa-ling pathway,which exerts a positive effect on the treatment of endometriosis(EMS).关键词
