四生丸加减方对原发免疫性血小板减少症小鼠Th17/Treg免疫失衡的调控机制研究
Regulatory Mechanism of Modified Sisheng Pill on Th17/Treg Immune Imbalance in Mice with Primary Immune Thrombocytopenia

Abstract
Objective:To explore the therapeutic characteristics and potential mechanism of Modified Sisheng Pill on mice with primary immune thrombocytopenia(ITP)based on Th17/Treg immune imbalance.Methods:The mice were randomly divided into 6 groups:blank control group(Control group),ITP group,prednisone group(Pre group),and low-,medium-,and high-dose of Sish-eng Pill groups(SSW-L group,SSW-M group,SSW-H group).Except for Control group,the ITP mouse model was established by im-munization in the other groups.After modeling,each administration group was given intragastric administration of the corresponding drug for 10 consecutive days.On the 11th day,blood samples,thymus,and spleen were collected.A hematology analyzer was used to determine the number of blood cells:platelet count(PLT),white blood cell(WBC)count,and red blood cell(RBC)count;the weights of thymus and spleen were measured to calculate the organ index;the number of total bone marrow megakaryocytes(MK)and platelet-producing megakaryocytes in the whole section was counted under a light microscope;ELISA was used to detect interleukin-6(IL-6),interleukin-17(IL-17),and transforming growth factor-β1(TGF-β1);flow cytometry to detect the percentage of helper T cells 17(Th17)and regulatory T cells(Treg)in spleen tissue;and Western blot to detect the protein expression of neurogenic gene Notch homolog 1(Notch1),retinoic acid-related orphan receptor-γt(ROR-γt),and forkhead box P3(Foxp3)in spleen tissue.Re-sults:Compared with Control group,ITP group showed decreases in PLT,RBC,MK,Foxp3 protein,TGF-β1,and Treg cells(P<0.05),while it showed increases in MK,IL-6 and IL-17,spleen index,thymus index,Th17 cells,Notch1 and ROR-γt(all P<0.05);Compared with ITP group,SSW-H group showed increases in PLT,RBC,MK,TGF-β1,Treg cells and Foxp3 protein(P<0.05),while decreases in spleen index,thymus index,MK,IL-17 and IL-6,Th17 cells,Notch1 and ROR-γt(P<0.05).Conclu-sion:The modified Sisheng Pill prescription can increase PLT in the peripheral blood of ITP mice,reduce the thymus index and spleen index,elevate the MK number in the bone marrow,and effectively regulate the Treg/Th17 immune balance.Its mechanism may be re-lated to the regulation of the Notch1/ROR-γt/Foxp3 signaling pathway.关键词
