Identification and functional characterization of DUX-like genes in early porcine embryos using Smart-seq2

Abstract
This study aimed to investigate DUX family genes in the early development of porcine embryos and their potential functions.Using Smart-seq2 single-embryo transcriptome sequencing combined with StringTie assembly,we performed a systematic analysis of 4-cell stage in vitro fertilized porcine embryos and successfully identified a DUX family gene,designated pDUXC,located at the terminal region of chromosome 17.Bioinformatic analyses suggested that the pDUXC protein contain two conserved HOX homeodomains and a proline-rich transcriptional activation region,along with distinct nuclear localization signals and the features of a transcription factor.Phylogenetic analysis revealed that pDUXC clusters closely with human DUX4,suggesting a high degree of evolutionary conservation between two species.Taken together,these findings suggest that pDUXC may participate in zygotic genome activation(ZGA)by activating ERVs,thereby driving the transcription of genes essential for early development.关键词
