摘要

Abstract

Objective:To explore the function of Total Eucomnmia Ulmoides Capsules on myocardial injury in spontaneously hypertensive rats(SHR)by regulating the reactive oxygen species(ROS)-mediated NLRP3 signaling pathway.Methods:A total of 8 male WKY rats were selected as the blank group,and 40 male rats were randomly divided into the model group,the positive(captopril,4.5 mg·kg-1)group,and low-dose group of Total Eucommia Ulmoides Capsules(92.5 mg·kg-1),the medium-dose group of Total Eucommia Ul-moides Capsules(185 mg·kg-1),and the high-dose group of Total Eucommia Ulmoides Capsules(370 mg·kg-1).Intragastric ad-ministration was performed for 8 weeks,while the blank group and model group were given equal volumes of deionized water via intra-gastric gavage.Caudal arterial blood pressure of rats in each group was measured before and after intervention.After having finished the administration,echocardiography was used to detect stroke volume(SV),cardiac output(CO),fractional shortening(FS),and ejection fraction(EF)of the rat'heart.HE staining was employed to observe myocardial structure.Flow cytometry was used to detect myocardial cell apoptosis and ROS levels.Western blot was adopted to determine the expression levels of Bcl-2,Bax,NLRP3,Caspase-1,and inter-leukin-1β(IL-1β)in myocardial tissue.Results:Compared with the blank group,the model group showed significantly increased blood pressure,myocardial cell apoptosis rate,and ROS levels(P<0.01),along with significantly decreased EF,FS,SV,and CO(P<0.05).In the myocardial tissue of the model group,the protein expression levels of Bax,NLRP3,Caspase-1,and IL-1β were significant-ly increased,while the protein expression level of Bcl-2 was significantly decreased.Compared with the model group,Total Eucommia Ulmoides Capsules significantly reduced blood pressure,myocardial cell apoptosis rate,and ROS levels,but remarkably increased EF,FS,SV,and CO levels,and significantly decreased the protein expression levels of Bax,NLRP3,Caspase-1,and IL-1β in myocardial tis-sue,but significantly increased the protein expression level of Bcl-2,and all the differences were statistically significant(P<0.05 or P<0.01).HE staining revealed that in the model group,myocardial cells were disarranged,with increased cell diameter,blurred stria-tions,and deformed and enlarged nuclei.Compared with the model group,the pathological changes in cardiac tissue of rats in the high-dose Total Eucommia Ulmoides Capsules group were significantly improved.Conclusion:Total Eucommia Ulmoides Capsules can im-prove cardiac function in SHR rats with hypertension by reducing ROS levels in myocardial cells,inhibiting cell apoptosis,and suppress-ing the activation of the NLRP3 pathway.
