Bioinformatics Analysis of Soybean NRPD1 Gene and Creation of Gene Editing Materials

Abstract
The NRPD1 gene encodes the largest subunit of RNA polymerase Ⅳ(Pol Ⅳ),which is a key component in the RNA-directed DNA methylation(RdDM)pathway.This pathway is one of the epigenetic regulatory mechanisms involving small RNAs in plants,primarily responsible for de novo DNA methylation in plants.Although the role of the NRPD1 gene has been extensively studied in Arabidopsis and some crops,there is a lack of reports regarding its function in soybeans.To uncover the function of NRPD1 in soybeans,this study first conducted sequence analysis of the NRPD1 gene in soybeans using bioinformatics methods.Subsequently,a knockout vector for the NRPD1 gene was constructed using CRISPR/Cas9 gene editing technology,followed by genetic transformation and se-lection of edited plants.Homologous gene searches in the soybean database revealed the presence of two copies of the NRPD1 gene in soybeans,named GmNRPD1a and GmNRPD1b.These two genes exhibit certain differences in their expression patterns across different tissues.The successful con-struction of the gene editing vector and acquisition of edited soybean plants provide crucial genetic materials for understanding the function of the NRPD1 gene in soybeans.This lays an important foun-dation for further exploration of the biological functions of NRPD1 in soybeans.关键词
