Bioinformatics Analysis of Soybean Dof Transcription Factor Family and Cloning of GmDof4 Genes Related to Oil Synthesis

Abstract
Dof-class transcription factors,a class of plant transcription factors with specific con-served domains,receive increasingly much academic attention for their important role in plant meta-bolic regulation,root development,gene defense regulation and other processes.Based on the previ-ous transcriptomic sequencing analysis of soybean hypolinolenic acid mutants,33 soybean Dof tran-scription factors with high expression level were selected,and the physicochemical properties,kin-ship and protein interaction analysis of the encoded proteins were performed;to further determine the function of Dof4 interacting with the acetyl-CoA carboxylase(accD)catalyzing oil synthesis,the plant expression vector of GmDof4 gene was cloned and constructed,and the promoter element and bind-ing site prediction were analyzed against the 2 000 bp sequence upstream of the start codon of Gm-Dof4 gene.The results showed that the 33 GmDof transcription factors all contained the specific mo-tifs CX2CX21CX2C and were unevenly distributed across 15 chromosomes in soybean;The proteins encoded by these transcription factors were predicted to be localized in the nucleus;Among the 28 predicted protein-protein interactions,43%of the Dof transcription factors were directly or indirectly closely related to insect resistance,cold resistance,drought resistance,salinity resistance and seed development in soybean,and only Dof4 showed a direct interaction relationship with key enzymes in oil synthesis;The full-length CDS sequence of the GmDof4 gene was obtained by RT-PCR to suc-cessfully construct the pCAMBIA3301-Dof4 plant overexpression vector.The results of promoter ele-ment and binding site analysis indicate that GmDof4 gene may function together with multiple key enzyme genes in catalytic oil synthesis to affect soybean oil synthesis,in order to lay the preliminary foundation for the regulation of seed oil metabolism by transcription factors in soybean Dof.关键词
