鸽Viperin基因克隆及生物信息学功能分析
Cloning and bioinformatic functional analysis of the pigeon Viperin gene

Abstract
To investigate the gene structure and predict the biological function of the pigeon endoplasmic reticulum-as-sociated,interferon-inducible antiviral protein(Viperin),nest PCR was used to obtain the pigeon Viperin coding se-quence(CDS)in this study.The sequence characteristics,structural domains,phylogenetic relationships,and potential functions were analyzed by bioinformatics methods.The results indicated that the pigeon Viperin CDS spanned 1 077 bp,encoding a 358 amino-acid protein classified as unstable.The Viperin lacked a signal peptide,exhibited higher hydrophi-licity in the central region,and potentially contained a N-terminal transmembrane domain.Secondary structure analysis of Viperin revealed a composition of 33%α-helices and 13%β-sheets.Tertiary structural modeling of Viperin further dem-onstrated a conserved barrel-shaped architecture characterized by a-helices enveloping β-sheets.Multisequence alignment analysis of Viperin indicated high conservation across species,except in the N-terminal region.Protein interaction net-work analysis of Viperin identified functional associations with key immune regulators,including CMPK2,IRF1,and TRAF6.Collectively,these results will provide a theoretical basis for functional studies of pigeon Viperin.关键词
