Abstract

The study aims to investigate the effect of adding Idebenone(IDE)to the semen extender on low-tempera-ture preservation of semen,and to provide a theoretical basis for optimizing the semen extender and improving the quality of semen after preservation.In this study,fresh semen was collected from 5 Hu sheep aged 2-3 years old with similar body weights and good health conditions.The semen that passed the test was mixed and diluted with different concentra-tion of IDE before being stored at low temperature.The qualified semen was mixed and diluted with diluents containing different concentrations of IDE[0 group(control group),5 μmol·L-1 group,10 μmnol·L-1group and 15 μmol·L-1 group]for cryopreservation,with three replicates in each group.We detected the changes in malondialdehyde(MDA)content on the 1st,3rd and 5th days,as well as sperm motility,vitality,plasma membrane integrity and acrosome integ-rity from 0 to 5 days.The results showed that with the extension of storage time,sperm motility,activity,and motility gradually decreased,and MDA content gradually increased.The motility and activity of the 10 μmriol·L-1 group from the 2nd to the 5th day were significantly higher than those of the other groups(P<0.05);the VSL of the control group from the 3rd to the 5th day,and the VCL and VAP from the 2nd to the 5th day were significantly lower than those of the 10 μmol·L-1 group(P<0.05);the plasma membrane and acrosome integrity rates of the 10 μmol·L-1 group throughout the process were significantly higher than those of the control group(P<0.05);the MDA content of the 10 μmol·L-1 group was also significantly lower than that of the other groups(P<0.05).The experimental results indicated that adding a certain proportion of IDE to the diluent could improve the quality of Hu sheep sperm,alleviate the damage to the sperm mem-brane,and achieve the effect of improving the quality of Hu sheep semen.When 10 μmriol·L-1 IDE was added to the cryopreservation diluent,the preservation effect of Hu sheep semen was the best.
