摘要

Abstract

The aim of the present study was to reveal the underlying molecular mechanism by which lncRNA JPX regu-lated the activity of rheumatoid arthritis fibroblast-like synoviocytes(RA-FLS).The reverse transcription-quantitative re-al-time polymerase chain reaction(RT-qPCR),Western blot,methylated RNA immunoprecipitation(MeRIP)assay,lu-ciferase reporter assay,RNA immunoprecipitation(RIP)assay,protein stability assay,and in vitro functional experi-ments were performed to explore the role and molecular mechanism of JPX in regulating RA-FLS activation.The results showed that JPX increased the N6-methyladenosine(m6A)modification in p21 mRNA,which increased its interaction with YTHDC2 to upregulate the expression of p21 protein in RA-FLS.Meanwhile,JPX further increased the enrichment of p21 protein in the cytoplasm of RA-FLS and subsequently promoted their proliferation.In summary,JPX can regulate RA-FLS proliferation through an m6 A-YTHDC2-p21 axis and the distribution of p21 protein in cells.JPX might be a no-vel target for RA diagnosis and treatment.
