Research advances in Mu-opioid receptor biased agonists
摘要
Abstract
Globally,approximately one-fifth of adults experience various pain-related issues,highlighting the critical need for effective pain treatment and management within the realm of global public health.Traditional opioid drugs achieve their analgesic effects by activating the Mu-opioid receptor in the body;however,their severe side effects have significantly restricted their clinical application.Recent studies indicate that the anal-gesic effects of opioids are mediated through the G protein signaling pathway,while certain side effects are as-sociated with the β-arrestin signaling pathway.Therefore,the design and development of Mu-opioid recep-tor-biased ligands have emerged as a promising strategy and focal point in analgesic drug research.This arti-cle provided a comprehensive review of the advancements in Mu-opioid receptor-biased ligand research,en-compassing synthetic small molecules and peptides with G protein bias,as well as natural compound-derived Mu-opioid receptor ligands,aiming to serve as a valuable reference for the advancement of G protein-biased opioid drugs.关键词
