Abstract
Objective To explore the correlation between plasma microRNA-939-5p(miR-939-5p),miR-365-3p with macular edema(ME)of retinal vein occlusion(RVO).Methods Prospective case-control study.98 patients with secondary ME of RVO admitted to our hospital from May 2021 to May 2023 were collected as the ME group,and 106 healthy individuals without ophthalmic diseases were included as the control group.Baseline data were collected.ELISA was applied to detect the expression levels of plasma miR-939-5p and miR-365-3p.According to the degree of ME,pa-tients were divided into mild group(n=28),moderate group(n=40),and severe group(n=30).The prognosis was fol-lowed up and patients were separated into the good visual prognosis group(n=67)and the poor visual prognosis group(n=31).Spearman's method was applied to analyze the correlation between plasma levels of miR-939-5p and miR-365-3p and the degree of secondary ME of RVO.The receiver operating characteristic(ROC)curve was applied to analyze the predic-tive value of plasma miR-939-5p and miR-365-3p for poor visual prognosis in patients with secondary ME of RVO.Logistic multivariate regression was applied to analyze the influencing factors of poor visual prognosis in patients with secondary ME of RVO.Results The plasma levels of miR-939-5p and miR-365-3p in the ME group decreased compared to the control group(all P<0.05).Compared with the mild group,the plasma levels of miR-939-5p and miR-365-3p in the moderate and severe groups decreased(all P<0.05).Compared with the moderate group,the plasma levels of miR-939-5p and miR-365-3p in the severe group decreased(all P<0.05).The levels of miR-939-5p and miR-365-3p in plasma were nega-tively correlated with the degree of secondary ME of RVO(r=-0.546,-0.463,all P<0.001).There were no statistically significant differences in the course of RVO,age,gender,smoking history,RVO type,body Mass Index(BMI),baseline central macular thickness(CMT),alcohol consumption history,and persistent macular edema between good visual prog-nosis group and poor visual prognosis group(all P>0.05).Compared with the good visual prognosis group,the proportion of severe edema increased in the poor visual prognosis group,while baseline best corrected visual acuity and plasma miR-939-5p and miR-365-3p levels decreased(all P<0.05).MiR-939-5p and miR-365-3p were protective factors for poor vi-sual prognosis in patients with secondary ME of RVO(all P<0.05).The AUC for predicting poor visual prognosis in pa-tients with secondary ME of RVO using miR-939-5p and miR-365-3p alone and in combination was 0.799,0.815,and 0.933,respectively.The probability of combined prediction of miR-939-5p and miR-365-3p was higher than that of single prediction(Z=3.135,3.036,P=0.002,0.002).Conclusions The plasma levels of miR-939-5p and miR-365-3p in pa-tients with secondary ME of RVO decrease,they are related to the disease severity and can serve as auxiliary indicators for predicting poor visual prognosis in patients.关键词
