摘要

Abstract

Taking tissue-cultured seedlings of apple rootstock G935 as the test material,the effects of different water contents and different concentrations of plant growth regulators on the ex vitro rooting effect of tissue-cultured seedlings when using vermiculite as the substrate were explored.The results showed that when the water content of vermiculite was 28%,the rooting rate of tissue-cultured seedlings reached 68.67%,with an average of 11 roots per seedling,the average length of primary lateral roots was 24.60 mm,and the average length of the longest root was 39.64 mm;all these indicators were higher than those when the water content of vermiculite was 8%,18%,24%,and 38%.On the basis of 28%vermiculite water content,dipping the base of seedlings in 4 000 mg/L IAA for 3 seconds achieved the best ex vitro rooting effect on tissue-cultured seedlings,which was better than that of IBA and NAA.After 30 days of cultivation in an artificial climate chamber,the rooting rate of tissue-cultured seedlings reached 97.1%,with an average of 15 roots per seedling,among which the length of primary lateral roots was 44.25 mm,and the length of the longest root was 60.47 mm.
