摘要

Abstract

[Objective]To investigate the developmental mechanism of lateral branch formation in Schima superba,a species of evergreen broad-leaved afforestation tree,and to identify branching-related genes.[Methods]This study conducted phenotypic observations on the lateral bud growth of Schima superba,measured endogenous hormone contents at three stages:dormant buds(S1),sprouting bud(S2),and young shoots(S3),and analyzed the expression patterns of differential genes using transcriptome sequen-cing.[Results]The lateral buds of Schima superba initiated and produced new branches primarily in early March.A period of"rapid growth"in lateral branch development was observed between March 19th and April 9th.The total number of shoots on the front segment(0~10 cm)of secondary branches was more than 10 times that of the rear segment(10~20 cm).The average number of branches from Fujian was sig-nificantly higher than that from Jiangxi and Zhejiang.The contents of GA3 and BL were significantly higher in S2 and S3 than in S1,with no significant difference between S2 and S3.The contents of JA and ZR/IAA were significantly higher in S3 than in S1 and S2,with no significant difference between S1 and S2.The transcriptome analysis revealed that a total of 152 differential genes were involved in hormone signal transduction pathways.WGCNA analysis identified three candidate genes related to JA and BL:MYC2(Ssu06G00023),BEH2(Ssu01G00918),and XTH23(Ssu07G01586).[Conclusion]Lateral branch growth of Schima superbafollows a"slow-fast-slow"pattern,with branching concentrated within the upper 0~10 cm of the branches.There are significant differences in the average number of branches among different origins.GA3 and BL play important roles in lateral bud germination,while JA and ZR/IAA maybe crucial regulators during the branching process of Schima superba.Three candidate genes related to JA and BR were identi-fied through transcriptome analysis,and protein interaction networks further validated their promoting roles in lateral bud germination and branching of Schima superba.
