Preliminary Research on the Initiation Culture System of"Hua jin"Camellia oleifera

摘要

Abstract

In order to establish an in vitro regeneration system for'Hua jin'Camellia oleifera,this study compared the parameters of different explants(leaves,stem segments,and floral organs),sterilizing time(6-13 min),and hormones(6-BA,PIC),and thus We then screened out the suiTab.combinations of explants,sterilization time and hormones for in vitro regeneration of'Hua jin'Camellia oleifera.The results showed that leaves and flower organs were the best explants for'Huajin'Camellia oleifera,and the optimal sterilization time was 8 min and 7 min,respectively,and the stem segments were not easy to regenerate.5.0 mg·L-1 6-BA+2.0 mg L-1 PIC was the optimal hormone combination for leaf regeneration,and 3.0 mg·L-1 6-BA+2.0 mg·L-1 PIC was was the best hormone combination for the regeneration of floral organs,and the proliferation of floral organ healing tissues was better after subculture.In conclusion,it can be seen that the floral organs and leaves are the best explants for the starter culture of'Hua jin'Camellia oleifera,and the combination of 6-BA and PIC is good for the induction of its healing tissues,and the healing tissues originated from the floral organ explants proliferated better and dedifferentiated thoroughly.
