摘要

Abstract

The study aims to establish a high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)method to determine the dissipation dynamics and final residues of 40%difenoconazole SC in Gentiana scabra Bunge,and to provide recommendations for its rational usage.Fresh root and dried root samples of Gentiana scabra Bunge were extracted with acetonitrile and water,followed by phase separation with sodium chloride.The acetonitrile phase was purified with PSA and C18 adsorbents,diluted with acetonitrile,and subsequently detected by HPLC-MS/MS.Within the standard calibration range of 0.2 to 20.0 ng/mL,the correlation coefficient for difenoconazole was r>0.999 0.After the pesticide application,the half-life of difenoconazole in the fresh roots of Gentiana scabra Bunge ranged from 6.0 to 6.9 days,with a maximum residue limit of 0.012 mg/kg at 21 days and 0.027 mg/kg at 14 days.In the dried roots,the half-life ranged from 5.6 to 8.0 days,with a maximum residue limit of 0.058 mg/kg at 21 days and 0.11 mg/kg at 14 days.The recommended application rate is 72 to 120 g a.i./ha,applied at the initial stage of the occurrence of Septoria leaf spot of Gentiana scabra Bunge.Pesticide treatment should be performed 2 to 3 times at 7-10 interval,with a pre-harvest interval of 14 days.
