达乌里秦艽组织培养技术研究
Research on the Tissue Culture Technology of Gentiana dahurica

Abstract
The exploration of the tissue culture technology of Gentiana dahurica aims to provide technical support for its large-scale development and utilization.With the leaves of G.dahurica seedlings as the materials,the effects of different treatments on their growth and differentiation were analyzed,and the regeneration system was investigated.The results demonstrated that the leaves of G.dahurica seedlings could be used to establish a plant regeneration system through tissue culture.The optimal condition for sterilizing the explants was 75%alcohol for 30 seconds,followed by 0.1%HgCl2 for four minutes.The optimal conditions for callus induction were MS+1.0 mg·L-1 NAA+1.0 mg·L-1 6-BA,with the highest induction rate reaching 93.03%,and the earliest time for callus induction was 23 days.The optimal conditions for adventitious bud in-duction were MS+0.8 mg·L-1 NAA+0.5 mg·L-1 6-BA,with an induction rate of 36.13%.The earliest time for bud formation was 20 days.The optimal condition for adventitious root induction was 1/2 MS+2.0 mg·L-1 IAA+0.5 mg·L-1 IBA,with an induction rate of 96.71%,an average of seven roots per plant,and an average maximum root length of 12.84 cm.The earliest time for rooting was ten days.The tissue-cultured seedlings hardened for ten days when the soil substrate was V(peat soil)∶V(perlite)=3∶1,and the transplanted seedlings grew well,with the best survival rate of 86.49%.This study established a tissue culture and plant regeneration system for G.dahurica,laying the foundation for its artificial rapid propagation and protection of germplasm resources.关键词
