Effect of the blood-activating and stasis-resolving mixture on the TGF-β1/Smad signaling pathway in rabbit knee osteoarthritis model

摘要

Abstract

Objective To investigate the regulatory effect of the blood-activating and stasis-resolving mixture on the TGF-β1/Smad signaling pathway in a rabbit model of knee osteoarthritis(KOA).Methods Thirty New Zealand rabbits were randomly divided into blank group,sham operation group,model group,Chinese herbal decoction group,and positive drug group.After successfully modelling by modified Hulth method,the rabbits were given gavage according to the groups for 4 weeks.4 weeks later,knee cartilage of rabbits was taken to observe the injury degree of rabbit articular cartilage,and the expression levels of PPAR,PARP1,TGF-β1,Smad2/3 and p-Smad2/3 proteins were detected.Results The model group exhibited significant articular cartilage damage,with Mankin scores significantly higher than those of the blank control and sham-operated groups(P<0.01).Expression levels of PPAR and PARP1 were upregulated,while protein levels of TGF-β1 and p-Smad2/3 were downregulated.Additionally,mRNA expression of TGF-β1,p-Smad2/3,and Col1A1 was reduced(P<0.01).In contrast,both the Chinese herbal compound group and the positive drug group showed significantly lower Mankin scores than the model group(P<0.01).Protein expression levels of TGF-β1 and p-Smad2/3 were higher than those in the model group,and mRNA expression of TGF-β1,p-Smad2/3,and Col1A1 was also elevated compared to the model group(P<0.01).Conclusion The blood-activating and stasis-resolving mixture can protect the articular cartilage and exert an intervention effect on KOA by regulating the TGF-β1/Smad signaling pathway in the rabbit knee osteoarthritis model and increasing the levels of proteins related to the TGF-β1/Smad signaling pathway.
