"扶正-通络-解毒"法干预大鼠毒瘀交阻型胃癌前病变作用机制
Mechanism of action of Fuzheng-Tongluo-Jiedu method in intervening precancerous lesions of gastric cancer in rats with toxin-stasis intermingling syndrome

Abstract
Objective:To investigate mechanism by Jianpi Yangyin Quyu formula regulates gastric mucosal epi-thelial-mesenchymal transition(EMT)via the IL-6/JAK2/STAT3 pathway in rats with precancerous lesions of gas-tric cancer(PLGC).Methods:A total of 58 SD rats were randomly divided into blank control group and model estab-lishment group.The PLGC rat model was induced using a multi-factor composite method.After successful modeling,the model establishment group were further divided into model group,folic acid group,and high-dose,medium-dose and low-dose Jianpi Yangyin Quyu formula formula groups(n=8 per group).After 8 weeks of treatment,HE stai-ning was used to observe pathological changes in gastric tissue.ELISA was performed to detect TNF-α and IL-6 lev-els.qPCR was used to measure mRNA expression of JAK2 and STAT3.Western blotting was employed to assess the protein expression of JAK2/p-JAK2 and STAT3/p-STAT3.Immunohistochemistry was applied to evaluate C-myc protein expression.Results:Jianpi Yangyin Quyu formula formula significantly ameliorated pathological manifesta-tions,including gastric mucosal atrophy,intestinal metaplasia,and dysplasia.It down regulated the expression of IL-6,TNF-α,JAK2,p-JAK2,STAT3,p-STAT3,and C-myc(P＜0.05),with the high-dose Jianpi Yangyin Quyu for-mula group exhibiting the most pronounced effects.Conclusion:Jianpi Yangyin Quyu formula formula alleviates PLGC by improving inflammatory microenvironment,inhibiting the activation of the JAK2/STAT3 pathway and C-myc expression,and suppressing the EMT process.关键词
