基于铁死亡代谢探讨电针心经对大鼠缺血性脑卒中的作用机制
Mechanism of electroacupuncture at heart meridian points on the ferroptosis metabolic pathway in rats with ischemic stroke

Abstract
Objective:To investigate effect of electroacupuncture at heart meridian points on the expression of proteins related to the ferroptosis iron metabolism pathway in neurons of rats with ischemic stroke.Methods:Male sprague-dawley(SD)rats were randomly divided into sham group,model group,Heart Meridian group,and non-acu-point group,with two time points first and seven days.The model was established using the modified longa filament method.The heart meridian group received EA at Jiquan Shaohai Lingdao and Shenmen on the affected limb,while the non-acupoint group received EA at non-acupoint points at the same segment level on the affected limb.Each ses-sion lasted 30 minutes daily.Neurological deficits were assessed using Longa scores;cerebral infarction volume was measured via TTC staining;histopathological changes were observed by HE staining;neuronal survival was evaluated via Nissl staining;Fe2 content and MDA in ischemic tissues was quantified by ELISA;and protein expression levels of nuclear factor erythroid 2-related factor 2 FTH,FTL and ferroportin 1 were analyzed by Western Blot.Results:Compared with the sham group,the model group showed increased neurological deficit scores,larger infarct volume,elevated Fe2 and MDA levels(P<0.05),and decreased neuronal count and protein expression of Nrf2,FTH1,FTL,and FPN1(P<0.05).Compared with the model group,the heart meridian group exhibited improved neurological function,reduced infarct volume,increased neuronal count,decreased Fe2 and MDA levels(P<0.05),and upregula-tion of Nrf2,FTH1,FTL,and FPN1 protein expression(P<0.05).The non-acupoint group showed no significant differences compared to the model group(P>0.05).Within the heart meridian group,comparison between the first and seven days showed further improvement in neurological function and infarct volume(P<0.05),along with in-creased expression of Nrf2,FTH1,FTL,and FPN1 proteins(P<0.05).Conclusion:Electroacupuntcture at heart me-ridian points can alleviate neurological impairment in rats,possibly through activating the Nrf2 pathway,regulating i-ron metabolism-related proteins,and inhibiting neuronal ferroptosis.关键词
