骨碎补柚皮苷干预退变软骨细胞IL-1β、TNF-α、MMP-3表达的研究
Drynaria fortunei Naringenin Intervening IL-1β,TNF-α,and MMP-3 Expres-sion in Degenerated Chondrocytes

Abstract
Objective To explore the mechanism of the Drynaria fortunei naringin in protecting articular cartilage by observing changes caused by Drynaria fortunei naringin intervention in inflammatory factors[interleukin-1 beta(IL-1β),tumor necrosis factor-α(TNF-α),and matrix metalloproteinase-3(MPP-3)]after IL-1 β-induced chondro-cyte degeneration.Methods Bilateral knee joints of New Zealand rabbits(4-week-old)were isolated aseptically.Then,an in vitro chondrocyte cultivation system was established by the type Ⅱ collagenase enzyme digestion method.The follow-ing groups were set up:a normal control group,and a degeneration model group induced by 10 ng/ml IL-1β,which was further divided into a blank serum group and a Drynaria fortunei naringenin-containing serum group(medicine-contai-ning serum group).After 48-hour treatments by distinct methods for each group,reverse transcription-polymerase chain reaction(RT-PCR)was employed to detect the IL-1 β,TNF-α,and MPP-3 messenger RNA(mRNA)expression in chondrocytes.Results The RT-PCR assay showed that the IL-1 β,TNF-α,and MPP-3 mRNA expression in IL-1 β-in-duced chondrocytes was reduced in the medicine-containing serum group.Conclusion IL-1β,TNF-α,and MPP-3 mRNA are overexpressed in IL-1β-induced degenerated chondrocytes.The mechanism of Drynaria fortunei naringenin in protec-ting articular cartilage may be related to the inhibition of IL-1 β,TNF-α,and MPP-3 mRNA expression in degenerated articular chondrocytes.关键词
