Abstract

25-Hydroxyvitamin D3(25(OH)VD3),as one of the main active forms of vitamin D3(VD3)in human body,provides high medicinal values.A recombinant strain WB600-pMA5-CYP109E1I85F-Fdx-FdR,which was previously constructed in our laboratory,could selectively hydroxylate C-25 of VD3 to produce 25(OH)VD3.Herein,our study was focused on further optimization of the fermentation process with this recombinant strain to convert VD3 to 25(OH)VD3.First,the types and concentrations of carbon sources,nitrogen sources,and metal ions in the culture medium were screened through single factor experiments and orthogonal experiments.As a result,the optimal medium components were peptone 50 g/L,yeast powder 50 g/L,glucose 30 g/L,and dipotassium hydrogen phosphate 9 g/L.Under the optimized conditions,the fermentation with this recombinant strain could convert 0.25 g/L VD3 to 10.01 mg/L 25(OH)VD3,which was 5.5 times more efficient than previous process.Then,the types and concentrations of cosolvents were investigated for such transformation to 25(OH)VD3.Delightfully,the addition of 3%(V/V)of DMSO to the medium could generate much higher concentration of product 25(OH)VD3(17.74 mg/L).Our research work demonstrated that optimization of the fermentation process with the recombinant strain could significantly increase the production of 25(OH)VD3.Further investigation of this transformation can be conducted at the fermentation tank level to improve the synthesis of 25(OH)VD3,presenting the great significance for the industrial application.
