摘要

Abstract

Objective To investigate the effect and its mechanism of early growth response factor 3(EGR3)on the migration and in-vasion of breast cancer cells based on the β-catenin/MMP pathway.Methods Real-time quantitative PCR(qPCR)and Western blot were used to detect EGR3 expression in different breast cancer cell lines(MDA-MB-231,MDA-MB-453,MCF-7 and HCC-1937)and human normal breast epithelial cell line MCF-10A.The cells with the lowest EGR3 expression were chosen as the research subjects.The cells were transfected with empty vector,EGR3 overexpression vector,and EGR3 overexpression vector plasmid+50 μg/mL β-catenin signaling pathway activator Wnt3a in empty group,EGR3 group,and EGR3+Wnt3a group,respectively.The MDA-MB-231 cells without transfection treatment were named as mock group.Scratch test and Transwell assays were used to detect cell migration and invasion;Western blot was used to detect the expressions of N-cadherin,E-cadherin,Vimentin,matrix metalloproteinase-7(MMP-7),MMP-9,β-catenin and glycogen synthase kinase-3β(GSK-3β).Results Compared with MCF-10A cells,EGR3 expres-sion in breast cancer cells was reduced(P<0.05),and the expression in MDA-MB-231 cells was the lowest.Compared with mock group,EGR3 expression was increased in EGR3 group(P<0.05),the cell migration and invasion abilities,and the levels of N-cad-herin,Vimentin,MMP-7,MMP-9,β-catenin,and GSK-3β were reduced(P<0.05),and E-cadherin level was increased(P<0.05).Compared with EGR3 group,EGR3 expression was reduced in EGR3+Wnt3a group(P<0.05),the cell migration and invasion abili-ties,and the levels of N-cadherin,Vimentin,MMP-7,MMP-9,β-catenin,and GSK-3β were increased(P<0.05),and E-cadherin level was reduced(P<0.05).Conclusion EGR3 can inhibit the migration and invasion of breast cancer cells by inhibiting the activa-tion of β-catenin/MMP pathway.
