Guava leaf extract alleviates islet cell injury in diabetic mice by inhibiting VEGF signaling pathway

摘要

Abstract

Objective To investigate the effects of Psidium guajava leaf extract on glucose metabolism and islet cell function in mice with type 2 diabetes mellitus(T2DM),and explore its potential mechanisms.Methods Twelve KK-Ay diabetic mice were randomly divided into T2DM group and Psidium guajava leaf extract group(1 638 mg/kg).After 8 weeks of intervention,glucose tolerance was assessed by oral glucose tolerance test(OGTT),and pancreatic tissue sections were examined using hematoxylin-eosin(HE)staining to evaluate islet histology.Network pharmacology and bioinformatics analyses were conducted to identify shared genes and signaling pathways between the active ingredients of the extract and T2DM.Key protein expressions were validated by immunohistochemical staining.Results Compared with T2DM group,mice in Psidium guajava leaf extract group showed significantly reduced fasting blood glucose and decreased blood glucose levels at all time points during OGTT(P<0.000 1).HE staining revealed obvious improvement in islet architecture.Quantitative immunohistochemistry showed significantly increased insulin expression in islets(P<0.000 1).Network pharma-cology analysis identified 128 potential targets of the active compounds of the extract,of which 85 overlapped with T2DM-related proteins,mainly enriched in the vascular endothelial growth factor(VEGF)signaling pathway,and other metabolic regulatory pathways such as the thyroid hormone pathway.Immunohistochemical staining further demonstrated significantly reduced expressions of VEGFA,p-P38,and AKT in Psidium guajava leaf extract group compared with T2DM group(P<0.01).Conclusion Psidium guajava leaf extract may alleviate islet cell injury and improve glucose metabolism in diabetic mice by inhibiting the VEGF signaling pathway.
