Effect of copper ion intervention on TGF-β1 expression and wound healing in keloid fibroblasts

Abstract
Objective To investigate the effect of copper ion(Cu2+)intervention on the expression of transforming growth factor-β1(TGF-β1)in human keloid fibroblasts(KFs)and its role in wound healing.Methods KFs were cultured in vitro and intervented with different concentrations of CuCl2 for 48 h.KFs were divided into control group(0 μmol/L),low concentration(10 μmol/L)group,medium concentration(20 μmol/L)group,and high concentration(40 μmol/L)group.TGF-β1 expression was measured by ELISA,cell proliferation was assessed using CCK-8 assay,cell cycle distribution was analyzed by flow cytometry,cell migration and scratch wound healing were evaluated by scratch assay,and cell invasion was examined using Transwell assay.Results Compared with con-trol group,TGF-β1 expression and cell survival rate showed no significant changes in low concentration group(P>0.05),and TGF-β1 expression and cell survival rate concentration-dependently decreased in medium and high concentration groups(P<0.05).Compared with control group,the wound healing rate and the cell migration and invasion abilities were significantly reduced in medium concen-tration group and high concentration group(P<0.05),and the changes of the above indicators were more significant in high concentra-tion group(P<0.05).Compared with control group,the G0/G1 phase cell arrest rate significantly increased in medium and high con-centration groups(P<0.05),while the S and G2/M phase cell arrest rates decreased(P<0.05).Conclusion Cu2+at moderate concen-trations(20-40 μmol/L)can reduce the TGF-β1 expression in KFs cells,inhibit cell proliferation,migration,and invasion,thereby slowing wound healing,which may be associated with cell cycle arrest.关键词
