摘要

Abstract

Objective To investigate the expression of miR-605-5p in hepatocellular carcinoma(HCC),and explore its effects and regulatory mechanisms on the proliferation,migration and invasion of human liver cancer cell line HepG2.Methods GEO data-bases,and chromogenic in situ hybridization(CISH)were used to analyze the expression of miR-605-5p in HCC tissues and adjacent normal tissues.The Kaplan-Meier plotter database was used to investigate the effect of miR-605-5p expression on the prognosis of HCC patients.HepG2 cells were divided into blank control group,miR-NC group(transfected with an empty vector)and miR-605-5p group(transfected with miR-605-5p expression vector),and the transfection efficiency was verified by qRT-PCR.Cell proliferation,migra-tion and invasion abilities were evaluated using cell counting kit-8(CCK-8),colony formation assay,wound healing assay and Tran-swell chamber assay,respectively,and the expressions of epithelial-mesenchymal transition(EMT)markers were detected by Western blot.The potential target genes regulated by miR-605-5p were predicted using TargerScan,miRWalk and miRDB databases,and ana-lyzed by Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.The hub genes of protein interaction network were screened using Cytoscape software.Results The expression level of miR-605-5p in HCC tissues was significantly downregulated compared to paracancerous normal tissues(P<0.05),and the low expression of miR-605 was associated with a poor prognosis in HCC patients.Compared to control group and miR-NC group,the proliferation,migration,and invasion abilities of HepG2 cells were reduced in miR-605-5p group(P<0.05),N-cadherin and Vimentin expressions decreased(P<0.05),and E-cadherin expression increased(P<0.05).KEGG enrichment analysis showed that the target genes regulated by miR-605-5p were mainly involved in signaling pathways regulating pluripotency of stem cells,tight junction,sphingolipid signaling pathway,phosphatidylinositol signaling system and so on.The key target genes of miR-605-5p,including KRAS,CREB1,MDM2,JAK2 and NANOG were obtained by protein interaction net-work analysis.Conclusion miR-605-5p is lowly expressed in HCC and exerts inhibitory effects on the proliferation,migration,and invasion of HepG2 cells,and may serve as a potential therapeutic target for HCC.
