摘要

Abstract

Objective To investigate the protective effects of Aucubin(AU)against cerebral ischemia-reperfusion(CIR)injury and elucidate its potential regulatory mechanisms.Methods Sprague-Dawley(SD)rats were randomly divided into four groups:control group,CIR group,CIR+AU 10 mg/kg group,and CIR+AU 20 mg/kg group.The CIR model was established using the middle cerebral artery occlusion(MCAO)method.The rats were intraperitoneally injected with the corresponding dose of AU daily for five consecutive days before modeling,and two days after modeling in CIR+AU 10 mg/kg group and CIR+AU 20 mg/kg group.The infarct volume was evaluated using triphenyltetrazolium chloride(TTC)staining,and the brain tissue damage was assessed by hematoxylin-eosin(HE)staining.Cell apoptosis was detected by TUNEL assay.Levels of reactive oxygen species(ROS),malondialdehyde(MDA),and super-oxide dismutase(SOD)were measured using specific assay kits.Levels of inflammatory cytokines including tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6),and IL-1β were quantified by ELISA.Protein expressions of AKT,glycogen synthase kinase-3 beta(GSK-3β),nuclear factor erythroid 2-related factor 2(Nrf2),heme oxygenase-1(HO-1),and quinone oxidoreductase 1(NQO-1)were determined by Western blot.Results Compared with control group,the cerebral infarct area,the brain tissue damage,the neu-ronal apoptosis,the oxidative stress,and inflammation levels significantly increased in CIR group(P<0.05).Compared with CIR group,the cerebral infarct area,the brain tissue damage,the apoptosis,the oxidative stress,and inflammation levels were signifi-cantly improved in CIR+AU 10 mg/kg group and CIR+AU 20 mg/kg group(P<0.05).Compared with control group,the levels of p-AKT and p-GSK-3β significantly decreased in CIR group(P<0.05),and the expressions of Nrf2,HO-1,and NQO1 proteins signifi-cantly increased(P<0.05).Compared with CIR group,p-AKT,p-GSK-3β,Nrf2,HO-1,and NQO1 protein expressions significantly increased in both CIR+AU 10 mg/kg group and CIR+AU 20 mg/kg group(P<0.05).All above indicators showed more significant effect in CIR+AU 20 mg/kg group than in CIR+AU 10 mg/kg group(P<0.05).Conclusion AU can exert significant protective effects against CIR injury,which may be closely related to the upregulation of AKT/GSK-3β/Nrf2 signaling pathway.
